The Yang lab at the University of Washington Department of Medicine/Cardiology, which collaborates closely with Dr. Chuck Murry's research group, is seeking 2 postdoctoral researchers with interests in using induced pluripotent stem cells (iPSCs) to model genetic cardiomyopathies due to sarcomeric mutations. This is a full-time position with a 12 months service period, with the possibility for yearly renewal dependent upon performance and continued funding.
The Yang lab has two major focuses. The first is to use patient-specific and gene-edited iPSC-derived cardiomyocytes to rapidly study multiple myosin heavy chain 7 variants and then phenotype them with high-throughput functional assays. A large number (on the order of thousands) of variants will be generated with cutting-edge gene-editing techniques in collaboration with our colleagues in Genome Sciences. As sarcomeric mutations act as poison peptides, the second focus of the lab is to understand the mechanisms that regulate the clearance of mutant sarcomeric proteins in order to leverage this knowledge for possible therapeutic potential. This involves innovative confocal microscopy techniques that the lab has developed to measure sarcomeric dynamics along with gain-of-function and loss-of-function experiments to study the role of key sarcomeric regulators in genetic cardiomyopathies.
These are highly collaborative projects that will expose the successful candidates to collaborators in Cardiology, Genome Sciences, Institute for Stem Cell and Regenerative Medicine (ISCRM), and Bioengineering. This position will be primarily supervised by Dr. Yang but will also have the opportunity to be co-mentored by Dr. Murry.
