The Stowers Institute for Medical Research has an opening for a Research Technician in the Screening core facility to perform high throughput production of lentivirus and adenovirus as well as perform cell-based viral-mediated screens.
Responsibilities include cloning cDNA, Cas9 and CRISPR guides into lentiviral and adenoviral vectors; high-throughput production of lentivirus and curation of viral libraries; optimization of viral production, viral titration, and MOI transduction of mammalian cells; utilization of robotic platforms for viral production, transduction of cells for screening and hitpicking for validation and counterscreens; generation of high titer bulk virus; and screening of stable mammalian cell lines transduced with virus.
In addition to excellent communication skills and the ability to work in a team-oriented environment, the successful candidate will have an attention to detail, strong leadership skills, ability to embrace working with technology/robots, mastery of tissue culture and transfection or transduction of mammalian or non-mammalian cells; prior experience with PCR, cloning, and other molecular biology techniques; and experience with lentiviral and/or adenoviral production.
Minimum requirements include an undergraduate degree in biology, microbiology, biochemistry or a related field with at least 4 to 7 years of related laboratory experience, or a Master’s degree in biology, microbiology, biochemistry or a related field with 2 to 3 years of related laboratory experience. Additional preferred skills include robotic experience with one or more robotic systems, programming and troubleshooting of robotic instrumentation, biological readouts in microtiter formats, assay development and screening in microtiter format, CRISPR assay development and screening; experience with cell-based reporter assay systems or screening small molecules; experience in high content imaging; bioinformatics experience; and proficiency with Microsoft Excel, Prism and Spotfire software.
